    Amsterdam's a great bicycling city, and parents commonly take their kids to school on bikes with child seats or trailers. Rent family bikes and explore the city's parks and museums (such as the Rijksmuseum and the Vondelpark).

    The Nemo science center is a must - great for kids, very hands-on. Can get busy in the weekend.

    Depending on how much time you have, I would also recommend getting out of the city, as usual the capital city is very different from the "real" Holland. Trains work well. I would look into Madurodam http://www.madurodam.nl

    I would pack a pic-nic and sit in the Vondelpark for a few hours. As mentioned Nemo is great and the new city library of Amsterdam is very nice and interesting. You can even make and send your own radio broadcast from there. Ask staff for other possibilities.

    The canal cruises are a classic and despite being a bit touristy a really nice way to see the city. Finally for some history, go to Anne Frank museum.

    Biking is definitely the thing to do with kids who know how to pedal without the extra wheels. We did it for the whole day There are great small bike rentals. Unfortunately many of them have problems with kids bike availability. The best bet is the most boring. Mac Bike, withe their premier outlet at the Centraal Station (at the eastern end) with heaps of bikes in all imaginable shapes an sizes available.
